ATPase Pumps
ATPase pumps acquire energy for ion translocation directly from the hydrolysis of ATP. The most prominent example of an ATPase pump is the Na+ pump (or more properly, the Na+/K+ ATPase pump), which is responsible for maintaining transmembrane concentration gradients for both Na+ and K+ (Figure 4.10A). The Na+ pump is a large integral membrane unit is responsible for ion translocation and spans the membrane ten times, with most of the molecule found on the cytoplasmic side, whereas the b subunit spans the membrane only once and is predominantly extracellular.

Ca2+ pumps are another class of ATPase pump (Figure 4.10B). Ca2+ pumps are an important mechanism for removing Ca2+ from cells. Two different types of Ca2+ pumps have been identified. One, called PMCA, is found on the plasma membrane; the other, termed SERCA, is used to store Ca2+ in the endoplasmic reticulum (see Chapter 7). The structure of SERCA (see Figure 4.10B) is remarkably similar to that of the Na+ pump, aside from having a binding site for Ca2+.

The crucial importance of the Na+ pump for brain function is evident from the fact that this transporter accounts for up to two-thirds of the brain’s total energy consumption. The Na+ pump of neurons was first discovered in the 1950s, when Richard Keynes at Cambridge University used radioactive Na+ to demonstrate the energy-dependent efflux of Na+ from squid giant axons. Keynes and his collaborators found that this efflux ceased when the axon’s supply of ATP was interrupted by treatment with metabolic poisons (Figure 4.11, point 4). Other conditions that lowered intracellular ATP also prevented Na+ efflux, proving that removal of intracellular Na+ requires cellular metabolism.

Further studies with radioactive K+ demonstrated that Na+ efflux is associated with the simultaneous ATP-dependent influx of K+. These energy-dependent movements of Na+ and K+ generate transmembrane gradients for both ions. The opposing fluxes of Na+ and K+ are operationally inseparable—removal of external K+ greatly reduces Na+ efflux (see Figure 4.11, point 2), and vice versa. Subsequent work by Jens Christian Skou in Denmark established that these fluxes of Na+ and K+ are due to an ATP-hydrolyzing Na+/ K+ pump. Quantitative studies indicate that Na+ and K+ are not pumped at identical rates: The rate of K+ influx is only about two-thirds that of Na+ efflux, indicating that the pump transports 2 K+ into the cell for every 3 Na+ that are removed.

The Na+ pump is thought to alternately shuttle Na+ and K+ across the membranes in a cycle fueled by binding of ATP and transfer of a phosphate group from ATP to the pump (Figure 4.12A). ATP binding promotes binding of intracellular Na+ and release of K+, while pump phosphorylation leads to extracellular release of Na+ and binding of K+. In between these two states of ion translocation are occluded states that prevent leakage of ions in the reverse direction, with subsequent hydrolysis of ATP leading to dissociation of ADP, which toggles the pump between accumulating intracellular K+ and removing intracellular Na+. While the precise mechanism of ion translocation is still being elucidated, many of the structuresinvolved have been identified (Figure 4.12B). Na+ and K+ alternately bind to sites in the interior of the pump, within the transmembrane domain of the a subunit. A nucleotide-binding domain is responsible for binding ATP, with hydrolysis of ATP transferring a phosphate onto a phosphorylation domain that subsequently changes the location of an actuator domain thought to regulate ion binding to the pump (see Figure 4.12B). This ATP-dependent structural rearrangement explains how the pump is able to move Na+ and K+ uphill, against their steep electrochemical gradients. Very similar structural changes are also thought to be involved in removal of cytoplasmic Ca2+ by the SERCA Ca2+ pump.

Ion Exchangers
The second class of active transporter does not use ATP directly but instead uses the electrochemical gradients of other ions as an energy source. Such ion exchangers carry one or more ions up their electrochemical gradient while simultaneously taking another ion (most often Na+) down its gradient. These transporters can be further subdivided into two types according to the direction of ion movement. As their name implies, antiporters exchange intracellular and extracellular ions. An example of an antiporter is the Na+/Ca2+ exchanger, which shares with the Ca2+ pump the important job of keeping intracellular Ca2+ concentrations low (Figure 4.13A). Another antiporter, the Na+/H+ exchanger, regulates intracellular pH (Figure 4.13B). Ion exchangers of the second type, the co-transporters, work by carrying multiple ions in the same direction. Two such co-transporters regulate intracellular Cl– concentration by translocating Cl– along with extracellular Na+ and/or K+; these are the Na+/K+/Cl– co-transporter, which transports Cl– along with Na+ and K+ into cells (Figure 4.13C), and the K+/Cl– co-transporter, which removes intracellular Cl– (Figure 4.13D) As you will see in Chapter 6, neurotransmitters are transported into synaptic terminals and glial cells via other co-transporters (Figure 4.13E). Although the electrochemical gradient of Na+ (or other counter-ions) is the proximate source of energy for both ion exchangers and co-transporters, these gradients ultimately depend on the hydrolysis of ATP by ATPase pumps such as the Na+ pump.

Summary
Ion channels and active transporters have complementary functions. The primary purpose of transporters is to generate transmembrane concentration gradients, which are then exploited by ion channels to generate electrical signals. The flow of ions through single open channels can be detected as tiny electrical currents, and the synchronous opening of many channels generates the macroscopic currents that produce action potentials and other electrical signals. A large number of ion channel genes creates channels with a correspondingly wide range of functional characteristics, thus allowing different types of neurons to have a remarkable spectrum of electrical properties. Voltage-gated ion channels are responsible for the voltage-dependent conductances that underlie the action potential. These channels are integral membrane proteinsthat open or close ion-selective pores in response to the membrane potential, allowing specific ions to diffuse across the membrane. Voltage-gated channels have highly conserved structures that are responsible for features such as ion permeation and voltage sensing, as well as the features that specify ion selectivity and sensitivity to neurotoxins. Other types of channels have specialized structures that enable detection of chemical signals, such as neurotransmitters or second messengers, or other types of signals such as heat or mechanical distortion of the plasma membrane. Active transporter proteins are quite different from ion channels because they move ions against a concentration gradient. The energy required for ion translocation is provided either by the hydrolysis of ATP or by the electrochemical gradient of other ions, such as Na+. The Na+ pump produces and maintains the transmembrane gradients of Na+ and K+ by ATP-dependent phosphorylation of the pump, which causes structural changes that enable ion movement. Other transporters, both ATPase pumps and co-transporters, are responsible for the electrochemical gradients for other physiologically important ions, including Cl–, Ca2+, and H+. Together, transporters and channels provide a comprehensive and satisfying molecular explanation for the ability of neurons to generate electrical signals.
